Overview

Workpackage 1 - USP New technologies & Glycoprotein applications
The focus in this work package is to address new technologies for an intensified/continuous culture process. This includes the development of Process Analytical Technology (PAT) for monitoring and control of perfusion processes, cell separation by centrifuge to support process intensification, and omics which is used as support for process and cell characterization.

Workpackage 2 - USP Model & Control
The focus in this work package is to develop methodologies for modeling, control and optimization of perfusion processes. Machine learning techniques is combined with domain knowledge and heterogeneous data sources such as metabolite concentrations and transcriptomics for high fidelity modeling. The models form the backbone for robust optimization and control algorithms developed for medium optimization and process control.

Workpackage 3 - DSP for Biologics & Gene therapy
The focus in this work package is to adapt and further develop the technology previously developed in AdBIOPRO 1 for downstream processing of biologics, particularly for antibodies, now switching the focus to gene therapy. The work package is focused on adeno-associated virus (AAV) and lenti virus (LV).

Workpackage 4 - USP AAV & Lentivirus
This work package focuses on the production of viral vectors for gene therapy using HEK293 mammalian cells. In particular, the manufacturing of adeno-associated- (AAV) and lentiviral (LV) vectors is addressed. A goal is to intensify the culture processes, using an approach amenable to large-scale application. Omics approaches are also used to characterize the production cells to optimize them and the culturing conditions.

Work package 5 - Cell Therapy
In this work package, the manufacturing process for the production of human cells as product is addressed, including the production of immune cells, mesenchymal stem cells and pluripotent stem cells. This includes the development of new tools or approaches for advanced Process Analytical Technology for monitoring and/or control, and omics is used for a better understanding of the cells.
