Work package 4 - USP AAV & lentivirus
Work package 4 focuses on the production of viral vectors for gene therapy using HEK293 mammalian cells. In particular, the manufacturing of adeno-associated- (AAV) and lentiviral (LV) vectors is addressed. A goal is to intensify the culture processes, using an approach amenable to large-scale application. For this, we focus on the improvement of the transfection process and ways to support ultra-high cell density cultures, using, for example, perfusion-mode reactors. Moreover, omics approaches are used to characterize the production cells to optimize them and the culturing conditions. WP4 addresses both cell and vector engineering for the production of lentiviral vectors. One target application for the vectors is the efficient generation CAR-NK cells.
